支原体是最小的可体外生长的微生物,可长期与宿主细胞共同生长,不产生急性细胞毒作用,在实验中容易被忽视。目前在细胞培养过程中有70%以上都存在支原体污染的情况。细胞受到支原体污染后不仅会影响细胞内某些基因的表达,也会在一定程度上影响细胞的生长状态。支原体污染的来源包括工作环境的污染、操作者本身的污染(一些支原体在人体是正常菌群)、培养基的污染、污染支原体的细胞造成的交叉污染、实验器材带来的污染和用来制备细胞的原始组织或器官的污染。
在细胞培养过程中应当定期做支原体检测,万一细胞已受支原体污染时,最佳的处理原则为将细胞高压灭菌后丢弃,以免污染其它洁净的细胞株。但是若受污染的细胞比较珍贵,必须去除支原体污染,则可以通过混合使用抗生素以达到目的。支原体没有细胞壁的,传统的抗生素一般对其无效。培养细胞支原体污染虽然不对细胞产生急性毒性作用,但可使细胞性状发生改变而影响细胞脂质体的转染和病毒对靶细胞的感染。包装细胞与靶细胞支原体污染的有效清除能明显改善病毒包装时的转染效率及靶细胞感染效率。
